I baked a two-tiered graduation cake, covered in fondant. When I put the top tier on, even with supports and a separator board, the bottom layer of fondant wrinkled on the bottom a bit. What can I do to prevent the wrinkling?

I will say the support was a bit short! A tiny bit of smashing and the wrinkles will form. Even an 8th of an inch can make a difference here!
