I am planning on making a toy story (Jessie) themed cake. I need the cake for 100 people. I thought about making a 12 x 4, then 8x6. Can this be done. I am still new to cake decorating and dont know how I need to support this cake. Can any body please give me any advise. It will be greatly appreciated.

Of course it can be done... stacking a 12 x 8. Are you asking if you can do it?

My advice: support your cake like all tiered cakes should be supported. Here's a Wilton page with instructions...
