At least a week for fondant, if you want them really hard. Semi-hard, a couple of days. Gumpaste dries a lot faster.

How hard do you want them? Do you want the recipients to be able to eat the flowers? If so, then they should be made and put on the cupcakes as close to display time as possible.

If you want them to pick the flowers off, then you can let them harden as long as you want, but as said before, fondant takes longer to dry than gumpaste.
