more info please ..... I'm not sure what kind of design you're going for with 'homemade' separators. Do you mean pillars or spacers?
If you're looking for spacer ideas, here's a cake I did this weekend: http://cakecentral.com/modules.php?name=gallery&file=displayimage&pid=1492272 (I hate the cake, btw, so overlook how crappy it looks). The separator between the tiers is just 5 pieces of cardboards cut to size, taped together, then wrapped with black ribbon. I used the width of the ribbon to determine how high to make the spacers.
For a pillar look, you might try small glasses, or building blocks glued together. Both of these will require some kind of support system underneath, though.
