You can paint whenever. 
As for the ratio, it depends on how much color you need. I put a little color in a small bowl and use a medicine dropper to add the vodka a little at a time until I get the right paint consistency. It should be too thick and goopy and should not be very runny--this is the same for mixing dusts as well. HTH
