Wedding Cake With Flowers Between Tiers

I have been asked to do a wedding cake to feed 100 people, covered in fondant, with red roses in between floating tiers.

From reading other posts, I have decided to use my normal SPS support, but use the extenders on the legs to add an extra 3" space - I'll fill this space with the roses.

The bride has asked me to provide the flowers (I would charge a fee, of course), and I have looked around online at wholesale places to purchase them.

My question is, how many roses do I need to fill the space?

The cake will be 12", flowers, 9", flowers, 6".

that is going to depend on the size of the roses, & how open they are. You might try a mock-up using cake pans & artificial flowers to get an idea.
