I would like to ask if there is any guide line for how much butter i need if I want a 4" to 5 " tall and 3 layer cake for a 6" 8" 10" round pan?.

How much butter?? It depends on your recipe. Figure out how much is used in each batch of batter then how many batters you need for those sizes of pans. Usually one batch of cake batter is used in a 10"x2" round pan. And about 1 batch in the other 2 pans. Then to get a 4" tall cake you have to make 2 of each size pan. So from my calculations you will be making 4 batches of batter. BTW: each cake 4" tall is called a 'tier' not a layer.
