AI have a three tiered cake with strawberry filling, buttercream icing and covered in marshmallow fondant. The strawberry filling was made from frozen strawberries, sugar, water and cornstarch. The filling set perfectly. My question is, does the cake have to be refrigerated or can it be left out overnight?

AIt doesn't seem like you have any perishable fillings, slit should be fine out overnight. If it has fresh strawberries or cream then you will have to refrigerate it.

ANo worries. Since you cooked them through. the sugar will preserve them, like jam. if you just threw them in without cooking them that would be another story.

It really depends on the weather where you are.  If it is HOT, like here in CA I suggest it be frig'ed :)  It also will transport better.
