I have fondant lips that I am going to put onto a 12x18 cake that will be decorated with white buttercream. Won't the color from the lips run onto the icing? I did word cutouts from the same fondant for the cake message and I used Wilton cake gel colors. Also, right now as they are drying I have cornstarched Saran Wrap under them positioned so they look like
3-D puffy lips but will they stay that way once they are on the cake and the Saran Wrap is taken out from under them?

You shouild not have any problem. The color should not run. Yes, they should dry as they are on the plastic wrap so they should be the same once placed on the cake.
